Graphite Bio doses first patient with GPH101 for sickle cell disease

Graphite Bio (NASDAQ:GRPH) is trading 2.1% higher after the company said it had finished dosing the first patient with its drug, GPH101, now called nulabeglogene autogedtemcel, in the company’s Phase 1/2 trial to treat people with sickle cell disease (SCD). The drug, Nula-celm is a gene editing therapy designed to directly correct the genetic mutation that causes SCD. "We believe nula-cel could be a definitive cure for sickle cell disease, with the potential to address all complications associated with this life-threatening disease. We look forward to reporting initial proof-of-concept data from the CEDAR trial in mid-2023," the company said. The trial will be conducted in about 15 patients with severe SCD. The trial is currently enrolling patients at multiple sites in the United States.

Shares of Graphite Bio are down some 85% from their June 2021 IPO pricing, as its first-ever clinical trial for the treatment of sickle cell disease experienced a six-month delay. The company’s potentially curative technology builds on CRISPR Therapeutics by pasting a DNA template into the target gene after it has been cut. With no actionable data likely forthcoming until mid-2023 but trading at a significant discount to cash, the recent insider buying merited a deeper dive. Graphite Bio: An Important Stock To Track In The Gene Editing Space

Graphite Bio Initiated the phase 1/2 CEDAR trial using GPH101 for a potential curative treatment of patients with severe sickle-cell disease. First patient in phase 1/2 CEDAR trial will be dosed in the 1st half of 2022 and the first set of results are expected in the 2nd half of 2022. It is estimated that the Sickle Cell Disease market is estimated to be worth $5.5 billion by 2023. If proof of concept achieved for GPH101 in Sickle-cell disease, then other indications to be tackled are Gaucher Disease and XSCID; IND filings for one of these not anticipated until mid-2023.

Graphite Bio: Assessing Prospects Amid Doubts Raised On Safety Of Crispr

Graphite Bio is a clinical-stage development biotech making use of CRISPR-based gene-editing therapy to develop a cure for sickle cell disease. Now, a biotechnology journal has published an adverse report on the safety of CRISPR-Cas9. As a result, the shares dipped, but have recovered partly. I make sense of the implications by going deeper into the report and, based on my assessment, the biotech's therapy has a differentiated gene correction approach. Operating in a highly specialized medical field and combating a life-threatening disease, the company is a buy at current prices, but the road towards commercialization will be a long and tortuous one.
