Wntresearch Provides an Update on the Work in Progress to Safeguard the Interests of Shareholders and Company Values WntResearch provides an update on the work to secure company values following the negative initial outcome analysis of the phase 2 study NeoFox. The company's drug candidate Foxy-5 showed no effect on tumors and lymph nodes as evaluated by CT, which therefore does not justify a continuation of the study. In order to safeguard the interests of the shareholders, the Board of Directors and management are working intensively on several parallel tracks where a goal is to complete a merger or a reverse acquisition. The Phase 2 clinical study NeoFox aimed to establish proof-of-concept for the drug candidate Foxy-5 for the treatment of colon cancer. The study included a total of 27 patients before it was stopped, including 16 patients on Foxy-5, and 11 patients in the control group, as previously announced. The initial result reading showed a favorable safety profile for Foxy-5, but without significant clinical benefit. The reason for the early analysis was to support the continued funding of the NeoFox study. However, no clinically relevant effects were seen on any of the study's crucial endpoints, which could be assessed in 15 out of 16 Foxy-5 patients, neither at half dose (n=3) nor at full dose (n=12). These endpoints were analyzed by CT scanning and included assessment of tumor size, as well as the number and size of lymph nodes, before and after treatment with Foxy-5. The exploratory endpoints included vascular and perineural invasion and were evaluated by tissue analysis of the surgically removed cancer tissue. In these analyses, the two groups are compared with each other. However, the number of patients who could be evaluated in each group was so limited (7 controls, 10 patients with full dose Foxy-5 and 3 patients with half dose Foxy-5) that it is impossible to draw any conclusiongs from the exploratory endpoints. To justify continued investments in the development of Foxy-5 for this indication, it would have been necessary to demonstrate large effects by CT already at this initial evaluation. The Company is of the view that the results achieved significantly reduce the possibility to raise additional money via future option programs or to attract a partner who would be willing to finance the extensive development program that wouldbe necessary for taking the drug canditade towards a future market approval. As a result, WntResearch decided to discontinue the study and has started to reduce the cost base and to safeguard the values that remain in the company. Discussions are ongoing with other companies regarding the possibility of carrying out a merger, reverse acquisition or similar. In parallel, the Board of Directors and management of WntResearch are also evaluating opportunities to realize the remaining value of the drug substance and the patent portfolio, and broadening the scope to pharmaceutical companies interested in the WNT signaling pathway both inside and outside oncology. WntResearch Presents Ad H Holes Observations from the NeoFox Study at ESMO GI Wnt Research AB (publ) announced that a poster on the previously communicated positive ad hoc observations from the ongoing clinical phase 2 study NeoFox, will be presented at ESMO Gastrointestinal Cancer Congress (ESMO GI), the leading European congress for gastrointestinal cancer held between June 26-29 in Munich, Germany. The poster presented at ESMO GI details ad hoc observations from 110 patients in the ongoing NeoFox study, indicating that a three-week pre-operative treatment with Foxy-5 rapidly reduces local tumour spread of colon cancer. Additionally, the poster includes new data on tumour (T)-staging. NeoFox is an open, randomized phase 2 trial in patients with stage II/III colon cancer that compares pre-operative treatment withFoxy-5 to an untreated control group. The results in brief: Pre-operative treatment with FoxY-5 was associated with an excellent safety profile. Historicopathological examination of surgically removed tissue suggested an effect of Foxy-5 on the local spread of the primary tumors within 3-4 weeks of treatment: Fewer patients had perineural invasion of cancer cells; Fewer patients had venous invasion of cancer cells; Foxy-5 was associated with TN(M) downstaging. The significance of histologic findings in a neoadjuvant setting in colon cancer is supported by an indirect comparison with neoadjuvant chemotherapy (FOLFOX). The title of the poster is: "Positive ad hoc results from neoadjuvant Foxy-5 treatment of colon cancer patients in the ongoing phase 2 NeoFox study." WntResearch has redesigned the study following the positive observations to focus on the early pre-operative effects in stage III colon cancer and plans to present initial efficacy data towards the year-end 2024. WntResearch AB Continues to the Next Step in the Phase 2 Study Neofox with the Highest Evaluated Dose of Foxy-5 WntResearch announced that the company, after a very fast patient recruitment, has been able to determine that the highest evaluated dose (8 mg/kg) of the drug candidate Foxy-5 will be used in the next step of the proof of concept study NeoFox. Approximately 80 patients with colon cancer will be treated in the second part of the study and an initial efficacy analysis is planned as early as the end of 2024. Previously reported ad hoc observations in patients with colon cancer indicate that the company's drug candidate Foxy-5 has a tumor-inhibiting effect already after three weeks of treatment. Based on the positive observations with Foxy-5, as well as the drug candidate's very favorable safety profile, the study plan for the clinical phase 2 study NeoFox has been optimized with the goal of being able to show early effects of Foxy-5. An important part of this work has been the dose-finding stage of the study, which has now been completed after very rapid patient recruitment. The results show that it is safe to proceed with the highest evaluated dose, 8 mg/kg, in the next part of the study. The dose selection has been approved by the study's Safety Committee. The Phase 2 clinical trial NeoFox aims to establish proof of concept for the drug candidate Foxy-5 in the treatment of colon cancer. Patients in the study receive the first dose of Foxy-5 after diagnosis and are then treated for three weeks until the primary tumor is surgically removed. WntResearch Continues to the Second Dose Group in Clinical Proof of Concept Study of Foxy-5 WntResearch announced that the safety evaluation from the first cohort in the dose-finding part of the proof-of-concept study NeoFox, support continued evaluation of the drug candidate Foxy-5 at an additional higher dose. After the most appropriate dose has been defined in the spring, approximately 80 patients with colon cancer will be treated. An initial efficacy analysis is planned based on data from 20 patients in active treatment and 20 patients in the control group. The Phase 2 clinical trial NeoFox aims to establish proof of concept for the drug candidate Foxy-5 in the treatment of colon cancer. Patients in the study receive the first treatment with Foxy-5 after diagnosis and are then treated for at least three weeks until the primary tumor is surgically removed. Two higher doses than previously used are now being evaluated, and then the most suitable dose will be used in the treatment of further 80 patients. WntResearch is planning an initial analysis of the data after 20 patients in the Foxy-5 group and 20 patients in the control group have had their tumor removed. The purpose of this initial analysis is to evaluate the effect of Foxy-5 and at the same time assess whether the planned number of a total of 80 patients is large enough for the study as a whole to demonstrate the desired effect. Final study results are expected in 2025. The NeoFox study was originally intended to include a two-year follow-up period of the patients. Positive ad hoc observations generated in the study enabled the design to be adjusted, which provides great benefits in terms of time and cost savings.
